coil check

I replaced 1 coil a few years ago. I have been experiencing rough performance,lack of power on hills etc and it threw a code for no 2 cylinder .... Can I use a multimeter to
check the integrity of the coil packs..??? I am installing new plugs in a few days..

You can't really test the performance as far as I know but to rule out the obvious, are the plugs new ish?And you confirmed the injector isn't clogging/clogged?
