Looking to buy a 2008 Yaris Sr - good price, good condition, but does have just over 100k miles. Is this a problem? Can I expect things to get expensive quite quickly??

In short, no. There are people with over half a million miles on their Yaris'.
I think the longer answer is that even a car with 10k miles on it could be a bad buy if it was involved in accidents and driven like it was stolen for every one of it's miles.
If you say it appears in good condition, chances are it'll be with you for the long haul with only typical maintenance/repairs to worry about.
I just purchased a 2007 Yaris LB with 167k miles. Drove it 120+ miles home after I bought it, changed the front brakes, oil and air filter. Runs like a charm,everything works, except the e-brake. Some rust.
I previously owned a 2002 Echo Sedan with 130k+ miles. Similar experience. These are very stout cars, mechanically at least, primarily because of there simplicity. Just be wary of rust or prior accidents.

100k...it probably has the original brake pads.
550k on mine.

How's the paint? On my black 08 the entire roof is peeling off. Primer and all down to the metal which is causing it to rust.

I got mine in Aug, with 156,300, and it's solid. Only had to change oil, filters, and clean the A/C drain.
Also, for some reason, the front seat belt sensor was unplugged, saved a few bucks from this site's tips! Was going to get it looked at by dealer.
